Cryptosporidium analyses usually result in a highincidence of nondetectable samples because of low concentrationsof oocysts and low recovery efficiencies inenvironmental water, suggesting greater volumes ofwater samples. However, there are limitations in analyzinghigh sample volumes, such as low filter performanceand limited volume-processing capabilities when usingUS Environmental Protection Agency method 1622.This study provides guidelines for using water turbiditymeasurements to determine optimum samplevolumes for the detection of Cryptosporidium oocystsusing method 1622. The purpose of collecting largervolumes of water samples is not necessarily to increasethe effective sample volume. The optimum samplevolumes suggested in this article can minimize themethod's limitations and consequently achieve betterrecovery of oocysts in water. Use of the optimum-volumesstrategy suggested in this study would savesample filtration time, enhance filter performance, andoffset the poor recovery efficiency of oocysts that typicallyresults from higher volumes of water and highturbidity.
