The evaluation of new methods for the detection of protozoan parasites in water samples is fraught with problems. The whole procedure contains many potential sources of variation. In this study, several of the potential sources of variation were evaluated to determine their potential effect on the performance data which are generated during method comparisons. The potential source of variation examined included: the age of oocysts; the viability of oocysts; methods of counting seeding doses; and, the volume of pellets examined.
