A new Cryptosporidium detection system, the ProNetic(TM) System, has been developed which utilizes immunomagnetic separation (IMS) to purify oocysts from water samples. Visualization and enumeration of organisms is achieved by immunofluorescent staining and observation using an epifluorescence microscope. Recovery of oocysts from spiked samples was examined using the ProNetic(TM) IMS System alone, and in combination with Gelman Envirochek(TM) filtration. The results indicate that the ProNetic(TM) System provides improved recovery and reproducibility when compared to results previously reported by others using the USEPA method outlined in the Information Collection Rule (ICR). Using the Envirocheck(TM) filter in combination with the ProNetic(TM) System, allowed the analyst to detect the presence of Cryptosporidium in a 10 liter water sample in approximately four hours.
