Peptide nucleic acid probes targeted against the 16S ribosomal RNA molecule were designed and used in in situ hybridization experiments for the specific detection of E. coli. The species specific probe was labelled with biotin which was subsequently detected with streptavidin horseradish-peroxidase and the Tyramide signal amplification system. E. coli cells were concentrated by membrane filtration prior to hybridization and the labelled cells detected by an automated laser scanning device called the ChemScan. Preliminary tests suggest that the detection of E. coli and other phylogenetically defined bacteria can be achieved within 3 hours.
