The objective of this paper was to determine whether Cryptosporidium oocysts retained infectivity following recovery from environmental water samples using US Environmental Protection Agency's Draft Method 1622. In-vitro excystation demonstrated that oocyst viability was unaffected by the recovery procedures, and infectivity was monitored using in-vitro cell culture combined with molecular detection methods. The results clearly demonstrated that oocysts recovered by Draft Method 1622 retained their infectivity for the two human cell lines tested (Caco-2 and HCT-8). A protocol was developed that allowed the in-vitro infectivity assay to be performed directly on oocysts recovered using Draft Method 1622, without modification of the procedure. Consequently, Draft Method 1622 may be used for source water or finished water monitoring when infectivity information is required, leading to improved risk assessments.
