Genotypic identification has been widely recognized as an effective tool in theidentification and characterization of microbial and infectious disease organisms. Amplification-based assays have been developed for many types of bacterial identification assays, including bacterial species discrimination and antimicrobial resistance determination. Molecular methods for bacterial identification consist of either target or signal amplification. As with all amplification assays, multiplexing either target or signal amplification is difficult. The authors have recently described a multiplex assay on an electronic microchip array which was capable of multiplex amplification of 10 targets simultaneously, with little decrease in amplification efficiency. In this paper, the authors describe a novel in situ amplification assay which simultaneously multiplex amplifies 10 different DNA targets. The paper also shows the reliance of this novel technology on Nanogen, Inc.'s proprietary microelectronic chip array for amplification and identification of genomic bacterial DNA targets.
