Most analytical methods for quantification of pathogens and other such discrete particles in water arehighly uncertain. This paper addresses one important source of variability in such methods:the recovery efficiency, or proportion of particles observed. A model for uncertainty in these methodsproposed by Nahrstedt and Gimbel1 is summarized to illustrate the experimental and statistical contextof this work. A Monte Carlo simulation method is then developed and applied to evaluate the quality ofexperimental designs to quantify variability in recovery. In particular, this method is applied to developpreliminary guidelines pertaining to the scope, the number of samples, and the number of particles ineach sample to be used in recovery studies. Finally, two commonly applied techniques for theestimation of recovery parameters are presented and contrasted. Includes 7 references, tables, figures.
