This powerpoint presentation begins by providing background information on: coliform bacteria, water quality and safety;E. coli and coliforms, indicators of potential faecal contamination; absence of indicator bacteria, does not guarantee absence of pathogens; and,eight commonly used methods in Australian water testinglaboratories for the detection of coliform bacteria. The objective of this study was to evaluate the eight enumeration methods routinely used in watertesting laboratories for the detection of coliform bacteria in watersamples using a precise number of bacteria. Materials and methods included:BioBall™ vials containing freeze-dried E. coli and E. aerogenescultures; testing all methods 10 times with each BioBall™ culture; the mean and standard deviation of all readings was calculated; and,three different batches of BioBall™ were used. Conclusions were that:for methods enumerating 1 ml samples (Petrifilm and SPC), Petrifilmand PCA-P produced the most accurate and consistent results;for methods enumerating 100 ml samples (MF, DST and MPN),Colilert™ produced the most consistent results;MF, Colisure™ and MPN produced results with greater variability thanfirst expected; and,BioBall™ is a very suitable quality control for evaluating the efficiencyof water testing procedures.
